PREPARATION
OF 3-AMINOPROPYLTRIETHOXYSILANE (APS, AES, APES or SILANE) TREATED SLIDES
FOR ADHESION OF DNA (e.g. IN MICROARRAYS) OR TISSUE ADHESION
- Clean slides
following protocol 5.1 in Practical In situ Hybridization (i.e. Soak
in Chromic Acid solution for at least 4 hours, wash well in running
water, then in distilled water and ethanol, oven dry).
(In fact, slides following
this pretreatment bind chromosomes, extended DNA fibres (DNA fibers),
and DNA in solution for microarrays quite effectively.)
- Immerse
slides in a freshly prepared 2% solution for 3-aminopropyltriethoxysilane
(e.g. Sigma A3648) in dry acetone for 30 seconds. Note 1.
- Wash twice
in distilled water
- Dry overnight
at 37C and store coated slides at room temperature (indefinitely; we
have used 1 year without problems).
Note 1:
Use a glass Coplin jar reserved exclusively for this purpose. The solution
does not keep, but many slides can be coated at one time. Use in Fume
Hood.
